Eliminate Metal Ion Interference in Oligo LC/MS

Metal ions are ubiquitous in regular HPLC columns and instruments that contain stainless steel components. Iron ions can cause a wide range of problems from analyte adsorption to sample oxidation. In this application note, we show that improved oligonucleotide peak shape and area can be achieved by switching to an Agilent Ultra Inert Altura Oligo HPH-C18 column.

 

Comparative LC/MS data demonstrate enhanced recovery, sensitivity, and reproducibility, while reducing metal adduct formation that complicates mass spectra.

 

The study also highlights workflow considerations to minimize alkali metal contamination, enabling cleaner spectra and more confident identification of impurities and low-level components.
